Development and Characterization of Expressed Sequence-tagged Simple Sequence Repeat Markers for Denphal-type Dendrobium Cultivars and Transferability to Dendrobium Species

Denphal-type Dendrobium is the most popular orchid for cut flower and potted plant trade. To improve commercial traits, many novel cultivars have been produced through hybridization by breeders. However, genetic relationship of unclear, thus hindering progress breeding programs. Therefore, development molecular markers encouraged to identify different cultivars. In this study, based on transcriptome database ‘Red Bull’, polymorphisms expressed sequence tag-derived simple repeat (EST-SSR) were developed from 100 pairs EST-SSRs that randomly selected EST-SSR database. The 42 was analyzed according EST-SSRs. Then, transferability performing a analysis 40 species. results showed total 5174 potential identified with 4486 unigene sequences, 5289 primer successfully designed. Of EST-SSRs, 86 expected polymerase chain reaction products primary screening, 58 fragment size, 20 polymorphisms. Furthermore, dendrogram at distance 0.15, collected grouped into five clusters three major two minor clusters; all these had same characters each cluster. 18 among species polymorphic.
